Neonatal haemolytic disease was diagnosed in a Bos indicus cross bred beef herd in south-east Queensland. The evidence suggested that this was due to B. argentina vaccination for babesiosis while non-pregnant or less than one month pregnant. The cows received 5 or 6 vaccinations during their breeding life, which indicated that antibody levels can persist for long periods. The autopsy, histopathological and haematological findings indicated that the mechanism was disseminated intra-vascular coagulation which occurred in calves dying within 24 hours of birth. During mouse embryogenesis, reversion of imprinted X chromosome inactivation in the pluripotent inner cell mass of the female blastocyst is initiated by the repression of Xist from the paternal X chromosome. Here we report that key factors supporting pluripotency-Nanog, Oct3/4, and Sox2-bind within Xist intron 1 in undifferentiated embryonic stem (ES) cells. Whereas Nanog null ES cells display a reversible and moderate up-regulation of Xist in the absence of any apparent modification of Oct3/4 and Sox2 binding, the drastic release of all three factors from Xist intron 1 triggers rapid ectopic accumulation of Xist RNA. We conclude that the three main genetic factors underlying pluripotency cooperate to repress Xist and thus couple X inactivation reprogramming to the control of pluripotency during embryogenesis. 相似文献

Polysaccharides are highly heat-sensitive macromolecules, so high temperature treatments are greatly destructive and cause considerable damage, such as a great decrease in both viscosity and molecular weight of the polymer. The technical feasibility of the production of exopolysaccharides by deep-sea bacteria Vibrio diabolicus and Alteromonas infernus was previously demonstrated using a bioproduct manufacturing process. The objective of this study was to determine which sterilization method, other than heat sterilization, was the most appropriate for these marine exopolysaccharides and was in accordance with bioprocess engineering requirements. Chemical sterilization using low-temperature ethylene oxide and a mixture of ionized gases (plasmas) was compared to the sterilization methods using gamma and beta radiations. The changes to both the physical and chemical properties of the sterilized exopolysaccharides were analyzed. The use of ethylene oxide can be recommended for the sterilization of polysaccharides as a weak effect on both rheological and structural properties was observed. This low-temperature gas sterilizing process is very efficient, giving a good Sterility Assurance Level (SAL), and is also well suited to large-scale compound manufacturing in the pharmaceutical industry. 相似文献

Pulsed-field gel electrophoresis (PFGE) analysis was developed and compared with random amplified polymorphic DNA (RAPD) method to type 18 Mycoplasma synoviae (MS) strains. All analysed strains were typeable by RAPD but only 89% of MS strains were typeable by PFGE because of DNA degradation. The discriminatory power of RAPD was greater than that of PFGE but the two techniques had a discriminatory index superior to 0.95, the threshold value for interpreting typing results with confidence. The in vitro, in ovo and in vivo reproducibility of both typing techniques was 100%. However, the interpretation of RAPD patterns was complicated because of inconsistent band intensity. Biopolymers produced by marine organisms can offer useful tools for regenerative medicine. Particularly, HE800 exopolysaccharide (HE800 EPS) secreted by a deep-sea hydrothermal bacterium displays an interesting glycosaminoglycan-like feature resembling hyaluronan. Previous studies demonstrated its effectiveness to enhance in vivo bone regeneration and to support osteoblastic cell metabolism in culture. Thus, in order to assess the usefulness of this high-molecular weight polymer in tissue engineering and tissue repair, in vitro reconstructed connective tissues containing HE800 EPS were performed. We showed that this polysaccharide promotes both collagen structuring and extracellular matrix settle by dermal fibroblasts. Furthermore, from the native HE800 EPS, a low-molecular weight sulfated derivative (HE800 DROS) displaying chemical analogy with heparan-sulfate, was designed. Thus, it was demonstrated that HE800 DROS mimics some properties of heparan-sulfate, such as promotion of fibroblast proliferation and inhibition of matrix metalloproteinase (MMP) secretion. Therefore, we suggest that the HE800EPS family can be considered as an innovative biotechnological source of glycosaminoglycan-like compounds useful to design biomaterials and drugs for tissue engineering and repair. 相似文献

Citrus black spot (CBS), caused by Phyllosticta citricarpa McAlp Van der Aa, was recently detected in southern Florida in the US. In addition to infected plant propagation materials, movement of infected citrus fruit poses a concern for potential spread of the disease out of the current quarantine zone, because lesions with pycnidia and conidia could develop after harvest. The conditions conducive for mycelial growth and development of pycnidia and conidia are not well known. Therefore, effects of temperature and relative humidity on growth and conidial production of P. citricarpa were determined and used as parameter inputs in CLIMEX to predict potential establishment of CBS in North America. Colony growth and conidial production in vitro were optimal at 27 °C, whereas there was no growth below 4 °C and above 37 °C. On fruit, lesion development and conidial production were observed at 4 °C, though at a low rate, indicating a greater versatility of the fungus on fruit. More full pycnidia were produced on the CBS lesions at 91 % RH compared to 84 %. Input parameters for CBS risk in CLIMEX obtained from literature, which reflected conditions for infection in spring/summer in Florida, predicted potential establishment in Florida but not in California. However, altering the parameter values to account for survival of the pathogen in leaf litter in winter predicted potential establishment in California as well as Florida. Thus, P. citricarpa could possibly establish beyond Florida if this organism is transported outside of the current quarantine zone to other citrus production areas. 相似文献
